Restriction digests of the clone give the following sizes (kb): EcoRI--3.1, 2.5, 0.6, 0.2; HindIII--5.5, 0.6, 0.3; SpeI--6.4. Plasmid expressing alkaline phosphatase. Foreign genes can be cloned upstream of the IRES/reporter gene, allowing production of a dicistronic messenger RNA in mammalian cells. IRES/reporter insert lacking polyA can be excised with NruI. |
